**552–02 is a small molecule epithelial sodium channel (ENaC) antagonist developed primarily by Parion Sciences and Kainos Medicines. It was designed as a potent, selective, and long-lasting ENaC blocker for inhalation delivery to the pulmonary system, aiming to enhance mucociliary clearance by expanding airway surface liquid (ASL) volume. The drug demonstrated up to two orders of magnitude greater potency than amiloride in blocking ENaC channels and was well tolerated in early clinical studies. Its main investigational indications included cystic fibrosis, xerostomia (dry mouth), chronic bronchitis, chronic obstructive pulmonary disease (COPD), radiation injuries, and bronchiectasis. Development of 552–02 has been discontinued for all known indications[2][4][5].**
