5D6 is a first-in-class peptide-linked monoclonal antibody targeting Neuropilin-1 (NRP1) designed to redirect pre-existing antiviral immunity toward solid tumors. The therapeutic strategy involves chemically conjugating or recombinantly fusing HLA-A2-restricted peptides (derived from viruses like CMV and SARS-CoV-2 or tumor-associated antigens) to the 5D6 antibody via a pH-sensitive cleavable linker. Upon binding to NRP1 on the surface of tumor cells, the 5D6 complex is rapidly internalized into endosomal and lysosomal compartments. This triggers a novel NRP1-mediated cross-presentation pathway where the peptide cargo is liberated and loaded onto MHC-I molecules for presentation on the tumor cell surface. This high-density presentation allows the tumor cells to be selectively recognized and eradicated by cognate CD8+ T cells, effectively repurposing abundant viral memory T cells for targeted oncology treatment.
