5D6-KLA is an experimental antibody-peptide conjugate (APC) designed to target B7-H3 (CD276), a transmembrane protein that is overexpressed in various solid tumors and plays a role in immune evasion. The construct consists of the 5D6 monoclonal antibody, which specifically binds to B7-H3, linked to a pro-apoptotic peptide known as (KLAKLAK)2 (KLA). Upon binding to B7-H3 on the surface of tumor cells, the conjugate is internalized via receptor-mediated endocytosis. Once inside the cell, the KLA peptide moiety disrupts the mitochondrial membrane, leading to the release of cytochrome c and the induction of apoptosis. This mechanism allows for the targeted delivery of a cytotoxic peptide payload directly to malignant cells, potentially reducing systemic toxicity compared to traditional chemotherapy. 5D6-KLA has shown preclinical efficacy in models of colorectal and breast cancer.
