5E5-28z CAR T cells are an investigational second-generation chimeric antigen receptor (CAR) T-cell therapy engineered to target the tumor-associated glycoform of Mucin 1 (TnMUC1). The construct utilizes the 5E5 single-chain variable fragment (scFv) for specific antigen recognition, coupled with a CD28 costimulatory domain and a CD3zeta signaling module. This design is intended to provide potent T-cell activation and rapid cytolysis of TnMUC1-positive tumor cells, which are prevalent in various adenocarcinomas such as pancreatic and breast cancers. In preclinical benchmarking, the 5E5-28z format is characterized by robust immediate effector functions and high cytokine output (including IFN-gamma and IL-2), though it is often used as a comparator for studying T-cell exhaustion and persistence relative to 4-1BB or CD2-based costimulatory designs.
