5F11-28Z CAR T cells are a novel anti-CD30 chimeric antigen receptor (CAR) T-cell therapy. This therapy involves genetically engineered T cells that express a CAR designed to target the CD30 protein, which is expressed on various lymphomas. The CAR construct incorporates a single-chain variable fragment (scFv) derived from the fully human antibody 5F11, along with CD28 hinge, transmembrane, and costimulatory domains, and a CD3ζ T cell activation domain. The therapy aims to activate T cells to recognize and eliminate CD30-expressing cancer cells. A Phase 1 clinical trial evaluated 5F11-28Z CAR T cells in patients with relapsed and refractory CD30-expressing lymphomas, including classical Hodgkin lymphoma and anaplastic large-cell lymphoma. However, the trial was halted early due to observed low efficacy and substantial toxicities.
