Overview

5G4 refers to two distinct monoclonal antibodies identified in biomedical research. In oncology, 5G4 is a novel monoclonal antibody targeting the Human Epidermal Growth Factor Receptor 2 (HER2), developed as a therapeutic candidate for HER2-positive cancers. It is characterized by high thermal stability (approximately 75°C) and a predominantly β-sheet secondary structure, exhibiting functional similarities to trastuzumab (Herceptin). In neurology, 5G4 (specifically Clone 5G4) is a highly selective monoclonal antibody that recognizes disease-associated, aggregated forms of alpha-synuclein, such as those found in Lewy bodies and Lewy neurites. This clone is widely utilized as a diagnostic and research reagent to identify pathological inclusions in synucleinopathies like Parkinson's disease and dementia with Lewy bodies, as it shows minimal reactivity to physiological monomeric alpha-synuclein.
