Drug intelligence / Profile preview
6,7-dichloro-2-methylsulfonyl-3-N-tert-butylaminoquinoxaline (DMB) is a small-molecule, orally active ago-allosteric modulator and agonist of the glucagon-like peptide-1 receptor (GLP-1R). Originally discovered by Novo Nordisk, DMB binds to an allosteric site on the GLP-1R, enhancing the binding affinity of endogenous GLP-1 while also directly stimulating cAMP production and glucose-dependent insulin secretion. In addition to its metabolic effects, DMB has been identified as a direct and selective gasdermin D (GSDMD) agonist that activates GSDMD pore formation and pyroptosis without requiring cleavage. This dual mechanism has led to preclinical research investigating DMB for the treatment of type 2 diabetes, obesity, transient focal cerebral ischemia (stroke), and as an immunotherapeutic agent to stimulate antitumor immunity in cancer.
