Drug intelligence / Profile preview
6-amino-6-deoxy paramylon (APM) is a cationized, aminated derivative of paramylon, a linear β-1,3-glucan polysaccharide sourced from the microalgae *Euglena gracilis*. Developed as a novel therapeutic agent to mimic the action of bile acid-binding resins, APM is designed for the treatment of obesity and type 2 diabetes. Unlike unmodified paramylon, APM effectively binds and adsorbs bile acids within the gastrointestinal tract, preventing their reabsorption. This sequestration promotes the fecal excretion of lipids and bile acids, which subsequently stimulates the liver to synthesize new bile acids from cholesterol via the upregulation of cholesterol 7 alpha-hydroxylase (CYP7A1) and downregulation of small heterodimer partner (SHP). Preclinical studies in diet-induced obesity mouse models have demonstrated that APM significantly reduces body weight gain, blood glucose levels, and serum LDL cholesterol without causing the gastrointestinal side effects typically associated with traditional bile acid sequestrants.
