Drug intelligence / Profile preview
6-aminocholestanol (6-AC) is a synthetic steroid derivative and antifungal agent that acts as an inhibitor of the eukaryotic translation initiation factor 4A (eIF4A). It has been investigated primarily as a selective inhibitor of the *Leishmania infantum* translation initiation factor 4A (LieIF4A) for the treatment of leishmaniasis. 6-aminocholestanol inhibits both the ATPase and helicase activities of eIF4A by perturbing ATP and RNA binding. It has demonstrated anti-leishmanial activity against both promastigotes and intracellular amastigotes of *Leishmania infantum*, validating LieIF4A as a potential drug target. The compound was developed through collaborative research involving the Institut Pasteur de Tunis, the Institut de Biologie Physico-Chimique, the Institut Pasteur de Paris, and the Centre Hospitalier Universitaire de Montpellier.
