Overview

**6-aminopicolamine (6AP)** is a novel, water-soluble, small molecule aminopyridine analog investigated for its potent and selective anti-metastatic activity in models of metastatic pancreatic ductal adenocarcinoma (PDAC)[1]. It acts as a prodrug, undergoing intracellular metabolism to bioactive derivatives which selectively inhibit **Phosphogluconate Dehydrogenase (PGD)**, a metabolic enzyme whose heightened activity is an adaptation supporting metastasis in a subset of pancreatic cancers. In preclinical models, 6AP reverses PGD-dependent malignant properties such as excessive glucose consumption and global histone hyperacetylation, with evidence for anti-metastatic efficacy and minimal toxicity in vivo. 6AP’s selectivity and lower off-target activity distinguish it from related compounds like 6-aminonicotinamide. Development is at a preclinical proof-of-concept stage for use in metastatic pancreatic cancer[1].
