This is a combination of three small molecule drugs—6-methylprednisolone (a synthetic glucocorticoid corticosteroid), colchicine (an anti-inflammatory alkaloid), and indomethacin (a nonsteroidal anti-inflammatory drug, NSAID). Each component has a distinct mechanism of action: - 6-methylprednisolone acts as an agonist at the glucocorticoid receptor, suppressing inflammation and immune responses. - Colchicine disrupts microtubule polymerization by binding to tubulin, inhibiting neutrophil motility and activity, thereby reducing inflammation. - Indomethacin inhibits cyclooxygenase enzymes (COX-1 and COX-2), blocking prostaglandin synthesis responsible for pain, fever, and inflammation. This combination may be considered in severe or refractory inflammatory conditions such as acute gout or relapsing polychondritis when monotherapy is insufficient. However, combining these agents increases the risk of gastrointestinal toxicity and other adverse effects; such regimens should only be used under close medical supervision[1][2][5].
