Drug intelligence / Profile preview
6-month adjuvant chemotherapy with 5-FU monotherapy is a chemotherapy regimen consisting of 6 months of adjuvant treatment with 5-fluorouracil (5-FU) as a single agent. 5-FU is a well-established small molecule antimetabolite and fluoropyrimidine that works by inhibiting thymidylate synthase, thereby disrupting DNA synthesis in rapidly dividing cancer cells. In the Phase 3 clinical trial NCT06609551, sponsored by Zhejiang Cancer Hospital, this regimen is being evaluated as a ctDNA-guided adjuvant chemotherapy strategy for elderly patients with high-risk stage II and stage III colon cancer post-surgery. Specifically, it is used as a de-escalation strategy in ctDNA-negative patients (compared to observation) and as a standard monotherapy in ctDNA-positive patients (compared to the more intensive XELOX regimen).
