This drug is a combination chemotherapy regimen known as TPDCV, consisting of five agents: 6-thioguanine, dibromodulcitol, lomustine (also called CCNU), procarbazine, and vincristine. Each component has a distinct mechanism of action: - **6-thioguanine** is a purine antagonist that incorporates into DNA and RNA, disrupting nucleic acid synthesis and function. - **Dibromodulcitol** is an alkylating agent that crosslinks DNA strands, inhibiting replication and transcription. - **Lomustine** (CCNU) is a nitrosourea alkylating agent causing DNA crosslinking and strand breaks. - **Procarbazine** is an alkylating agent that inhibits DNA, RNA, and protein synthesis through methylation of guanine residues in DNA. - **Vincristine** binds to tubulin to inhibit microtubule formation during mitosis. TPDCV has been studied primarily for the treatment of malignant brain tumors such as childhood anaplastic astrocytoma, glioblastoma multiforme, mixed anaplastic oligoastrocytomas, and anaplastic ependymoma. It has also been used in combination with radiotherapy. The regimen aims to exploit the synergistic effects of these drugs with different mechanisms to maximize antitumor activity[1][2].
