Overview

64Cu-DOTA-alendronate is a PET imaging radiopharmaceutical designed to target and visualize microcalcifications within breast tissue. The agent consists of alendronate, a bisphosphonate with high affinity for hydroxyapatite crystals, conjugated to a DOTA (1,4,7,10-tetraazacyclododecane-1,4,7,10-tetraacetic acid) chelator that coordinates the positron-emitting radioisotope copper-64. In breast cancer, microcalcifications are often associated with malignant lesions; 64Cu-DOTA-alendronate aims to differentiate malignant from benign calcifications based on the intensity of tracer uptake, potentially improving the specificity of breast cancer screening and diagnostic localization prior to surgery or biopsy.
