**64Cu-DOTA-cetuximab** is a radiopharmaceutical imaging agent consisting of cetuximab (a chimeric monoclonal antibody targeting epidermal growth factor receptor, EGFR), chemically conjugated to the chelator DOTA and radiolabeled with the positron-emitting isotope copper-64. Cetuximab blocks ligand binding at the EGFR, an oncogenic receptor often overexpressed in various solid tumors. Through radiolabeling, 64Cu-DOTA-cetuximab is used for positron emission tomography (PET) to non-invasively visualize and quantify EGFR expression in tumors. It binds with high affinity and specificity to EGFR-positive tumor cells and demonstrates receptor-mediated uptake, strongly correlating with EGFR expression[1][2][4][5]. The agent can be used to assess cetuximab accumulation in tumors and potentially predict therapeutic response in EGFR-expressing malignancies, notably including colorectal cancer[4].
