64Cu-DOTA-MAb159 is a radiolabeled monoclonal antibody used for PET imaging that specifically targets cell-surface glucose-regulated protein 78 (GRP78), a receptor overexpressed in many tumors and associated with growth, metastasis, and therapy resistance[1][3][6]. It consists of MAb159, a humanized anti-GRP78 monoclonal antibody, conjugated to the chelator DOTA and labeled with Copper-64, enabling specific and sensitive detection of GRP78-positive tumors in vivo for cancer imaging applications[1][3]. The mechanism hinges on antibody binding to GRP78, allowing imaging of tumors with high cell surface expression of this protein. Developed mainly for imaging pancreatic cancer, it has shown efficacy in imaging models of colorectal and prostate cancer as well[1]. MAb159 also has potential antitumor effects owing to inhibition of the PI3K signaling pathway mediated via GRP78 blockade[6].
