64Cu-NOTA-C60-PEG is a radiolabeled fullerene-based nanoparticle designed for use as a positron emission tomography (PET) imaging agent. The construct consists of a C60 fullerene core functionalized with 1,4,7-triazacyclononane-1,4,7-triacetic acid (NOTA) chelators, which facilitate stable coordination with the positron-emitting radioisotope copper-64 (64Cu). To enhance water solubility, biocompatibility, and pharmacokinetic properties, the nanoparticle surface is modified with polyethylene glycol (PEG) chains. In vivo studies indicate that this nanoplatform exhibits rapid renal clearance and small molecule-like behavior, which minimizes long-term retention and potential toxicity. While the base 64Cu-NOTA-C60-PEG construct serves as a non-targeted tracer or control, it is frequently utilized as a scaffold for the development of targeted diagnostic agents by conjugating it with specific ligands, such as cRGD peptides for imaging integrin αvβ3 expression in tumors like glioblastoma.
