Overview

64Cu-NOTA-FVIIai is a radiopharmaceutical PET imaging tracer designed to target and visualize tissue factor (TF) expression in tumors, particularly pancreatic adenocarcinoma. It consists of active site-inhibited coagulation factor VII (FVIIai), which serves as the natural ligand for TF, conjugated to the chelator 2-S-(4-isothiocyanatobenzyl)-1,4,7-triazacyclononane-1,4,7-triacetic acid (p-SCN-Bn-NOTA) and radiolabeled with the positron emitter copper-64 (64Cu). By utilizing an active site-inhibited version of FVII, the tracer retains high binding affinity for TF without triggering the coagulation cascade. It is being developed as a potential companion diagnostic to identify patients with high TF expression who may benefit from TF-targeted cancer therapies, as TF levels often correlate with advanced stage and poor survival in pancreatic cancer.
