Drug intelligence / Profile preview
64Cu-NOTA-TSHR-Fab is a preclinical radiopharmaceutical imaging agent designed for immuno-positron emission tomography (immuno-PET) imaging of thyroid-stimulating hormone receptor (TSHR) expression in thyroid cancer. It consists of the antigen-binding fragment (Fab) of the human anti-TSHR monoclonal antibody K1-70, conjugated to the bifunctional chelator 2-S-(4-isothiocyanatobenzyl)-1,4,7-triazacyclononane-1,4,7-triacetic acid (p-SCN-Bn-NOTA) and radiolabeled with the positron emitter copper-64 (64Cu). By targeting TSHR, which is highly expressed in thyroid cancer, 64Cu-NOTA-TSHR-Fab enables noninvasive detection of TSHR expression to help guide TSHR-targeted therapies. In preclinical mouse models of thyroid cancer, the radiotracer demonstrated rapid and superior TSHR-specific tumor accumulation, fast clearance, and low background signal compared to its single-chain variable fragment (scFv) counterpart.
