**64Cu-p-NH2-Bn-DOTA-hu14.18K322A** is a radiolabeled humanized monoclonal antibody developed for PET imaging of tumors expressing the ganglioside GD2. It consists of the anti-GD2 antibody hu14.18K322A site-specifically conjugated to DOTA (a chelator) via a p-aminobenzyl linker and labeled with the positron-emitting radioisotope copper-64 (^64Cu). The parent antibody, hu14.18K322A, is engineered with a K322A mutation to reduce complement activation and associated pain, yet preserves antibody-dependent cell-mediated cytotoxicity (ADCC). This radiotracer is primarily investigated to enable sensitive, specific PET imaging of neuroblastoma and melanoma, facilitating detection, monitoring, and assessment of anti-GD2 immunotherapies as well as guiding patient selection and therapy optimization[1][3].
