64Cu-Tamrada + 177Lu-Tamrada is a combination of nanomedicine radiopharmaceuticals designed to selectively target and deliver radioactive isotopes (copper-64 and lutetium-177) to tumor associated macrophages (TAMs) within solid tumors, particularly those resistant to immune checkpoint inhibitors. These agents do not require conventional targeting ligands and are engineered to maximize selectivity for TAMs and retention of the radiotherapeutic isotopes within the tumor microenvironment. Therapeutically, 177Lu-Tamrada delivers cytotoxic beta radiation to induce cell death and bystander killing of adjacent tumor cells, while 64Cu-Tamrada acts as a diagnostic agent for theranostic precision dosing. The combination enables imaging and targeted therapy (a theranostic approach) for cancers such as triple negative breast cancer that are resistant to standard therapies. Preclinical studies demonstrate significant tumor growth inhibition in models of anti-PD-1 resistant triple negative breast cancer.[1]
