68Ga-ABS011 is a radiopharmaceutical imaging agent designed for positron emission tomography/computed tomography (PET/CT) to non-invasively assess HER2 expression in cancer lesions throughout the body. It consists of a single domain antibody (sdAb) that specifically binds to the human epidermal growth factor receptor 2 (HER2), conjugated via a NOTA chelator and labeled with gallium-68, a short-lived positron-emitting radioisotope. The sdAb component enables rapid and specific targeting of HER2-positive tumor cells, while unbound tracer is quickly cleared from circulation, resulting in high contrast images with low background signal. This agent aims to overcome limitations of traditional biopsy-based HER2 testing by providing whole-body mapping of HER2 status and heterogeneity across all metastatic sites. Its primary clinical application is in metastatic breast cancer for guiding personalized anti-HER2 therapy decisions and monitoring treatment response[3][4][6].
