68Ga-DOTA-Manocept is a radiopharmaceutical imaging agent developed by Navidea Biopharmaceuticals for positron emission tomography (PET) imaging of tumor-associated macrophages (TAMs). The agent is constructed using Navidea's Manocept™ platform, which consists of a mannosylated amine dextran (MAD) backbone. This backbone is conjugated with DOTA (1,4,7,10-tetraazacyclododecane-1,4,7,10-tetraacetic acid) chelators to facilitate radiolabeling with the positron-emitting isotope gallium-68 (68Ga). The drug specifically targets the mannose receptor (CD206), a protein highly expressed on the surface of M2-polarized macrophages, which are prevalent in the tumor microenvironment and associated with immune suppression and poor prognosis. By binding to CD206, 68Ga-DOTA-Manocept enables the non-invasive visualization and quantification of macrophage density, providing a potential biomarker for cancer progression and response to immunotherapies. It is also being explored for imaging inflammatory conditions such as rheumatoid arthritis.
