Overview

68Ga-DOTA-mDesmo is an experimental radiopharmaceutical diagnostic tracer designed for positron emission tomography (PET) imaging of tumors that overexpress Desmoglein 3 (DSG3). Desmoglein 3 is a calcium-binding transmembrane glycoprotein and a member of the cadherin family of cell adhesion molecules, which is significantly upregulated in various squamous cell carcinomas (SCC), including head and neck squamous cell carcinoma (HNSCC), lung SCC, and cervical SCC. The tracer consists of a specific DSG3-binding monomeric peptide (mDesmo), derived from the binding domain of autoantibodies associated with pemphigus vulgaris, conjugated to the macrocyclic chelator DOTA and labeled with the positron-emitting radioisotope Gallium-68. By binding with high affinity to the extracellular domain of DSG3, 68Ga-DOTA-mDesmo enables the sensitive and specific localization of primary tumors and metastatic lesions, potentially improving the staging and monitoring of DSG3-positive malignancies.
