68Ga-Glu-urea-Lys-HBED-CC-DyLight800 is a dual-modality imaging agent designed for the preoperative detection and intraoperative navigation of prostate cancer. It is a small molecule conjugate derived from the established PET tracer 68Ga-PSMA-11, consisting of a PSMA-targeting Glu-urea-Lys motif, an HBED-CC chelator for Gallium-68 labeling, and the near-infrared fluorescent dye DyLight800. This dual-labeling approach allows for high-sensitivity PET imaging to identify tumors and metastases before surgery, followed by real-time fluorescence guidance to assist surgeons in achieving precise tumor resection. Preclinical studies have demonstrated that this compound exhibits significantly higher tumor uptake (approximately 3.2-fold) compared to 68Ga-PSMA-11, likely due to the increased lipophilicity provided by the DyLight800 fluorophore, while maintaining high binding affinity and specific internalization into PSMA-positive cells.
