Drug intelligence / Profile preview
68Ga-NK224 is an investigational, peptide-based PET diagnostic radiotracer targeting human programmed cell death ligand 1 (PD-L1). It consists of a cyclic peptide (NK224) derived from DK221, conjugated to a NOTA chelator, which allows for radiolabeling with gallium-68 (or alternatively fluorine-18). Developed by academic investigators at the First Affiliated Hospital of Xiamen University, 68Ga-NK224 is designed to noninvasively map and quantify whole-body PD-L1 expression, assess intratumoral and intertumoral heterogeneity, and monitor PD-L1 target occupancy during anti-PD-L1 immunotherapy. In clinical studies of patients with non-small cell lung cancer (NSCLC), 68Ga-NK224 PET/CT was well tolerated and demonstrated a strong correlation between tumor tracer uptake and PD-L1 expression levels determined by immunohistochemistry, offering a promising noninvasive alternative to single-site tissue biopsies for patient stratification and treatment monitoring.
