68Ga-NOTA-RCCB6 is a radiolabeled single-domain antibody-based immunoPET imaging agent designed to target CD70, a cell surface protein highly and specifically expressed in clear cell renal cell carcinoma (ccRCC). The molecule consists of the RCCB6 single-domain antibody conjugated with the chelator NOTA and labeled with gallium-68. Its mechanism involves binding to CD70 on tumor cells, enabling precise PET/CT imaging of primary tumors and metastases in ccRCC patients. Preclinical studies demonstrated high affinity for human CD70, rapid tumor visualization, and specificity confirmed by reduced uptake after CD70 blockade. In clinical pilot studies, 68Ga-NOTA-RCCB6 outperformed traditional 18F-FDG PET/CT in identifying CD70-positive ccRCC lesions. The agent is under investigation for its diagnostic accuracy in staging, therapeutic evaluation, and restaging of renal cell carcinoma[3][1][2][5].
