Gallium Ga 68-NOTA-Sgc8 is a radiopharmaceutical imaging agent composed of the DNA aptamer Sgc8, which is conjugated to the chelator NOTA and labeled with the positron-emitting isotope gallium-68. The Sgc8 aptamer specifically binds to protein tyrosine kinase 7 (PTK7), a cell surface receptor overexpressed in various human cancers, including colorectal and bladder cancer. Upon administration, the compound targets PTK7-expressing tumor cells, enabling their visualization through PET/CT or PET/MR imaging. Preclinical and first-in-human studies have demonstrated high specificity for PTK7, rapid blood clearance primarily via renal excretion, favorable safety profile without significant toxicity or genotoxicity at high doses, and effective tumor imaging capabilities. The drug is being developed as a diagnostic tool for early cancer detection and potentially for monitoring therapeutic response in PTK7-positive tumors[1][2][3][6][9].
