68Ga-THP-Trop2 VHH is an investigational ImmunoPET radiotracer designed for the non-invasive imaging of Trophoblast Cell-Surface Antigen 2 (Trop-2) expression in solid tumors. The agent consists of a Gallium-68 (^68Ga) radioisotope complexed with a tris(hydroxypyridinone) (THP) chelator, which is conjugated to a VHH (variable domain of heavy-chain only antibody), also known as a nanobody, specific for Trop-2. Trop-2 is a transmembrane glycoprotein frequently overexpressed in epithelial cancers and is a primary target for antibody-drug conjugates (ADCs). The use of a VHH allows for rapid blood clearance and high-contrast imaging due to its small size compared to full-length antibodies. The THP chelator facilitates rapid, one-step radiolabeling with Gallium-68 under mild conditions, making it suitable for clinical diagnostic workflows to identify patients who may benefit from Trop-2-targeted therapies.
