68Ga-XH06 (also designated as [68Ga]Ga-XH-06) is a radiopharmaceutical imaging agent developed by Union Hospital, Tongji Medical College, Huazhong University of Science and Technology for the diagnosis and staging of hepatocellular carcinoma (HCC). The agent consists of XH-06, a novel humanized single-chain variable fragment (scFv) that specifically targets glypican-3 (GPC3), a cell-surface proteoglycan highly overexpressed in most hepatocellular carcinomas but absent in healthy liver tissue. By radiolabeling the XH-06 scFv with the positron emitter gallium-68, the tracer enables high-resolution PET/CT or PET/MR imaging to detect GPC3-positive tumors. Clinical studies have demonstrated that 68Ga-XH06 possesses high sensitivity and specificity for HCC, including the ability to detect small lesions under one centimeter, while maintaining a favorable safety profile with minimal non-specific uptake outside of renal clearance pathways.
