Drug intelligence / Profile preview
6bK is a potent, selective, and physiologically active small-molecule macrocyclic inhibitor of insulin-degrading enzyme (IDE) with an IC50 of approximately 50 nM. Discovered by researchers at Harvard University through the screening of a DNA-templated macrocycle library, 6bK binds to an exosite of IDE rather than its catalytic active site, achieving over 1,000-fold selectivity over other metalloproteases. In animal models, 6bK regulates the levels of insulin, glucagon, and amylin, improving oral glucose tolerance and delaying gastric emptying. It is widely used as a research tool to study the physiological roles of IDE in type 2 diabetes, glucose homeostasis, and insulin clearance.
