6G5j-IR800CW is a **fluorescent imaging agent** consisting of the monoclonal antibody 6G5j conjugated to the near-infrared fluorescent dye IRDye800CW. The antibody component (6G5j) is a mouse IgG1 monoclonal antibody that binds to **carcinoembryonic antigen-related cell adhesion molecules (CEACAM)**, specifically CEACAM 1, 3, 5, 6, and 8. These molecules are highly expressed on colorectal cancer cells and other epithelial malignancies. The conjugate is developed primarily for **fluorescence-guided surgery** and **intraoperative imaging** of colorectal cancer, particularly **liver metastases**. The IRDye800CW component has an absorption maximum at 774 nm and emission maximum at 789 nm in PBS, enabling visualization in the near-infrared spectrum with minimal background interference. When administered intravenously, the conjugate specifically accumulates on CEACAM-positive tumors, allowing surgeons to visualize tumor margins and distinguish malignant tissue from normal tissue during surgical procedures. The imaging agent has been used in preclinical mouse models where it demonstrated effective tumor-to-liver ratios and enabled clear differentiation between tumor tissue and normal liver parenchyma when used in combination with ICG for dual-color imaging. The conjugate is prepared by coupling IRDye800CW-NHS ester to the 6G5j antibody through standard NHS ester chemistry, which forms amide bonds with lysine residues.
