7-valent pneumococcal-OMPC conjugate vaccine

The 7-valent pneumococcal-OMPC conjugate vaccine (PncOMPC) is an experimental biologic vaccine developed by Merck & Co. to prevent acute otitis media and pneumococcal infections in infants. It contains capsular polysaccharides from seven *Streptococcus pneumoniae* serotypes (4, 6B, 9V, 14, 18C, 19F, and 23F), each individually conjugated to the outer membrane protein complex (OMPC) of *Neisseria meningitidis* serogroup B, which serves as the carrier protein. The vaccine was notably evaluated in the randomized, double-blind Finnish Otitis Media (FinOM) Vaccine Trial conducted by the Finnish Institute for Health and Welfare (THL). In this trial, PncOMPC demonstrated approximately 56% efficacy against vaccine-type acute otitis media, showing a clinical efficacy profile comparable to the CRM197-conjugated vaccine (Prevnar), although it elicited functional antibodies with different kinetics. Despite its efficacy, this specific OMPC-conjugated formulation was not commercialized as a standalone product.
