70-28z-1A CAR T cells are an experimental chimeric antigen receptor (CAR) T-cell therapy designed to target CD70-expressing tumors, specifically renal cell carcinoma (RCC). This construct incorporates a second-generation CAR architecture (CD28 costimulatory domain and CD3ζ signaling domain) further modified with the inclusion of the pTα 1A domain derived from the pre-T cell receptor. The addition of the 1A domain is intended to mimic the proliferative burst seen during thymocyte β-selection, thereby enhancing T-cell expansion, cytokine production, and in vivo persistence while reducing exhaustion. Crucially, the 1A domain increases the antigen sensitivity of the CAR, allowing for the effective elimination of tumor cells with low target antigen density, a common mechanism of escape in solid tumors like RCC.
