70-28z CAR T cells are an experimental chimeric antigen receptor (CAR) T-cell therapy designed to target the CD70 antigen, which is frequently overexpressed in various hematologic and solid tumors, including renal cell carcinoma (RCC). The 28z designation indicates a second-generation CAR architecture that incorporates a CD28 costimulatory domain and a CD3ζ signaling domain to facilitate T-cell activation and proliferation upon antigen encounter. Developed through a collaboration between the Columbia Initiative in Cell Engineering and Therapy (CICET) and Memorial Sloan Kettering Cancer Center, these cells serve as a benchmark in studies evaluating novel CAR modifications, such as the addition of the pTα 1A domain (creating 70-28z-1A), to enhance antigen sensitivity and overcome tumor-mediated immune evasion. In preclinical models of renal cell carcinoma, 70-28z CAR T cells have been evaluated for their ability to clear tumors, though they often serve as a baseline comparison for more advanced constructs designed to handle low antigen density.
