Overview

7033 CAR-T is an experimental chimeric antigen receptor (CAR) T-cell therapy designed to overcome antigen heterogeneity and prevent tumor escape in acute myeloid leukemia (AML). The therapy utilizes a dual-targeting approach: the T cells are engineered with a CAR construct specific for CD70 and are further modified to secrete a bispecific T-cell engaging antibody molecule (TEAM) that targets CD33 and CD3. This "CAR-TEAM" platform allows the engineered cells to directly eliminate CD70-positive leukemia cells while simultaneously redirecting both the CAR-T cells and endogenous bystander T cells to attack CD33-positive leukemia cells. Preclinical studies published in 2024 and 2025 demonstrate that this approach achieves potent cytotoxicity across various AML cell lines and patient-derived samples, including those with deficient expression of a single antigen, without increasing tissue toxicity.
