740YP is a cell-permeable phosphopeptide that acts as a potent and specific activator of phosphoinositide 3-kinase (PI3K). It consists of a sequence derived from the platelet-derived growth factor receptor (PDGFR) phosphorylated at Tyr-740, coupled to a cell-penetrating peptide sequence (Antennapedia). 740YP works by binding to the SH2 domains of the p85 regulatory subunit of PI3K, thereby mimicking the natural activation process and stimulating the PI3K/Akt/mTOR signaling pathway. In clinical research led by the National Foundation for Fertility Research, 740YP is utilized in an 'In Vitro Activation' (IVA) protocol for women with primary ovarian insufficiency (POI) and infertility. In this procedure, ovarian tissue is treated ex vivo with 740YP and the PTEN inhibitor bpV(hopic) to awaken dormant primordial follicles before being autografted back into the patient to restore reproductive function.
