763 SOSIP is a rationally designed, recombinant HIV-1 envelope (Env) trimer immunogen developed by Imperial College London for the prevention of HIV infection. It is derived from a specific HIV-1 envelope sequence (isolate 763) identified in an individual who developed broad neutralizing antibody responses early in the course of infection. The SOSIP modification involves the introduction of a disulfide bond (SOS) and a proline mutation (IP) to stabilize the gp120-gp41 trimer in its native-like, pre-fusion conformation, which is essential for eliciting broadly neutralizing antibodies (bNAbs). In clinical trials, it is administered via intramuscular injection, often formulated with an adjuvant such as Monophosphoryl Lipid A (MPLA) in liposomes, to evaluate its ability to prime or boost the immune system's B and T cell repertoires as part of an experimental medicine strategy.
