Drug intelligence / Profile preview
7aaRGD interfering peptide is a synthetic 7-amino-acid peptide containing the arginine-glycine-aspartate (RGD) motif, developed by the Nencki Institute of Experimental Biology. It is designed to block the interaction between tumor-secreted ligands, such as secreted phosphoprotein 1 (SPP1, also known as osteopontin) and milk fat globule-EGF factor 8 (MFGE8), and their integrin receptors, specifically αvβ3 and αvβ5 integrins. By disrupting SPP1/integrin signaling, 7aaRGD prevents the reprogramming of glioma-associated microglia and macrophages (GAMs) into an immunosuppressive state, thereby reversing the "cold" tumor microenvironment. In preclinical models of glioblastoma and brain metastases (derived from breast and lung cancers), 7aaRGD has been shown to normalize tumor vasculature and, when combined with immune checkpoint inhibitors like anti-PD-1 antibodies, significantly reduce tumor growth and enhance anti-tumor immune responses.
