7C3 is a mouse monoclonal antibody (mAb) that acts as a potent agonist of the thyrotropin receptor (TSHR). Developed through hybridoma technology using TSHR289 protein immunization, 7C3 mimics the pathological activity of thyroid-stimulating autoantibodies (TSAb) characteristic of Graves' disease. Upon binding to the TSHR, 7C3 triggers the activation of both canonical and non-canonical NF-κB signaling pathways, leading to the upregulation of pro-inflammatory chemokines such as CCL2, CCL5, and CCL19 in thyrocytes. Experimental administration in murine models has demonstrated that 7C3 induces elevated serum thyroxine levels and thyroid follicular hyperplasia, establishing it as a significant tool for modeling Graves' disease and investigating the molecular mechanisms of autoimmune thyroid disorders.
