Overview

7DC2 is a monoclonal antibody that specifically targets the CD47 receptor, an immune checkpoint protein often overexpressed on the surface of various cancer cells. CD47 serves as a 'don't eat me' signal by interacting with SIRPα on macrophages, thereby inhibiting phagocytosis. 7DC2 binds to CD47 with a high affinity (0.56 nM) and is designed to block this inhibitory interaction, facilitating the immune-mediated clearance of tumor cells. In preclinical research, 7DC2 has been utilized as the targeting moiety for an antibody-drug conjugate (ADC) known as 7DC2-DM1, which carries the cytotoxic payload mertansine (DM1) via a non-cleavable linker. Studies in murine models of lung and colorectal cancer have evaluated its binding epitopes and antitumor efficacy, noting distinct conformational and penetration characteristics compared to other CD47-targeting clones.
