7DC4-DM1 is an experimental, non-cleavable CD47-targeting antibody-drug conjugate developed in an academic program at Fujian Normal University. It couples the anti-CD47 monoclonal antibody 7DC4 to the maytansinoid microtubule inhibitor DM1 through an SMCC-based non-cleavable linker. The antibody component binds CD47 and is intended to disrupt CD47-SIRPα antiphagocytic signaling, while intracellular delivery of DM1 inhibits tubulin function and induces tumor-cell cytotoxicity. In published preclinical work, 7DC4-DM1 showed high CD47-binding affinity, cytotoxic activity, and antitumor effects in murine colorectal cancer models, with lung cancer also evaluated experimentally.
