7E11.C5.3 is a murine monoclonal antibody (IgG1 kappa) that specifically targets the intracellular domain of the human prostate-specific membrane antigen (PSMA), a 100-kDa transmembrane glycoprotein. Developed by Cytogen Corporation, this antibody serves as the targeting component of the diagnostic imaging agent capromab pendetide (ProstaScint). When conjugated to the linker-chelator glycyl-tyrosyl-(N,-diethylenetriaminepentaacetic acid)-lysine (GYK-DTPA) and radiolabeled with Indium-111, it is used for radioimmunoscintigraphy to detect the extent of prostate cancer and identify soft tissue metastases. Because it targets an internal epitope of PSMA, it primarily binds to necrotic or apoptotic cells where the cell membrane is compromised, making it a standard tool for staging and monitoring recurrence in prostate cancer patients.
