Overview

8-Azaadenosine is a small molecule adenosine analog that acts as a potent inhibitor of Adenosine Deaminase Acting on RNA 1 (ADAR1). ADAR1 is an enzyme responsible for the conversion of adenosine to inosine (A-to-I) in double-stranded RNA transcripts, a process critical for regulating innate immune responses and RNA stability. In the context of pulmonary hypertension (PH) research, 8-azaadenosine has been used to demonstrate that ADAR1 deficiency promotes endothelial apoptosis and vascular remodeling by dysregulating the circadian gene Nocturnin (NOCT). While primarily used as a research tool to study RNA editing and innate immunity, its inhibition of ADAR1 has been shown to worsen indices of PH in experimental models, highlighting the protective role of ADAR1-mediated RNA editing in the pulmonary vasculature.
