Overview

**8-Azaguanine** is a synthetic purine analog and antimetabolite that closely resembles guanine and competes with it in cellular metabolism. It is incorporated into ribonucleic acids (RNA) via the enzyme hypoxanthine-guanosine phosphoribosyltransferase (HGPRT), forming azaGMP and disrupting purine nucleotide biosynthesis. This leads to inhibition of DNA/RNA synthesis and cell proliferation. Originally developed as an antitumor agent in the mid-twentieth century—one of the first purine analogs with demonstrated carcinostatic effects—it was studied for use in acute leukemia and other malignancies but is not currently approved for clinical use. Recent research also suggests immunomodulatory activity by potentiating natural killer (NK) cell cytotoxicity against blood cancer cells[1][2][3][4][7].
