Overview

8-bromo-9-ethyl-2-phenetoxyadenine is a synthetic small molecule designed as a high-affinity and selective antagonist of the **A2A adenosine receptor**. It belongs to the class of 2-phenethoxyadenine derivatives with substitution at the 8-position of the adenine ring, in this case with a bromine atom. This structural modification confers high affinity for the A2A adenosine receptor (K_i = 1.7 nM) and selectivity versus other adenosine receptor subtypes. In preclinical studies, it has shown potent antiparkinsonian effects in rat models of Parkinson’s disease, where it can revert haloperidol-induced catalepsy and potentiate levodopa-induced motor responses. The mechanism is through the antagonism of the A2A adenosine receptor, which functionally modulates dopaminergic signaling—an established therapeutic approach in Parkinson’s disease. The compound is a research tool and candidate molecule and is not commercially approved or marketed[1].
