Overview

8-ethoxy-2-phenethoxy-9-ethyladenine is a small molecule selective **adenosine A2A receptor antagonist**. It demonstrates high binding affinity (low nanomolar range) and functional antagonism at the human adenosine A2A receptor, while displaying good selectivity over other adenosine receptor subtypes. In animal models of Parkinson’s disease, it reverses haloperidol-induced catalepsy and potentiates the effects of L-dopa in 6-hydroxydopamine-lesioned rats, as well as reducing tremulous jaw movements in a model of parkinsonian tremor. This compound is proposed as a promising antiparkinsonian agent and nondopaminergic adjunct therapy, analogous in therapeutic approach to istradefylline, but distinct in chemical scaffold[1][3].
