Drug intelligence / Profile preview
8-pCPT-2-OMe-cAMP (also known as 007) is a synthetic, cell-permeable analog of cyclic adenosine monophosphate (cAMP) that acts as a potent and highly selective agonist for the exchange proteins directly activated by cAMP, EPAC1 (RAPGEF3) and EPAC2 (RAPGEF4). Unlike endogenous cAMP, 8-pCPT-2-OMe-cAMP selectively activates EPAC signaling pathways without activating protein kinase A (PKA) at typical working concentrations. It is widely utilized as a pharmacological research tool to elucidate PKA-independent cAMP signaling mechanisms in various physiological and pathological processes, including endothelial barrier function, insulin secretion, and cardiac remodeling. In preclinical models, pharmacological activation of EPAC1 with 8-pCPT-2-OMe-cAMP has demonstrated protective effects against the progression of glomerulonephritis and chronic kidney disease by promoting cellular energy adaptations and a glycolytic metabolic shift in podocytes.
