Drug intelligence / Profile preview
826827 (also known as humAb 826827) is a fully human monoclonal antibody that targets the Apical Membrane Antigen 1 (AMA1) of *Plasmodium vivax* (PvAMA1). PvAMA1 is an essential invasion protein utilized by both sporozoites during hepatocyte invasion and merozoites during reticulocyte invasion. 826827 binds to highly conserved residues within the hydrophobic groove of PvAMA1, blocking its interaction with its native receptor, Rhoptry Neck Protein 2 (RON2). This dual-stage inhibition prevents both pre-erythrocytic (liver stage) and blood-stage infections. Developed collaboratively by academic and research institutions including Case Western Reserve University, the Walter and Eliza Hall Institute of Medical Research, and the Burnet Institute, 826827 has demonstrated potent strain-transcending inhibitory activity in vitro and significantly reduced liver parasite burden in humanized mouse models in vivo.
