858 Therapeutics is developing a preclinical small molecule inhibitor targeting ADAR1 (adenosine deaminase acting on RNA 1) for the treatment of solid tumors. ADAR1 is an RNA-editing enzyme that converts adenosine to inosine in double-stranded RNA (dsRNA), a process that prevents the activation of innate immune sensors such as MDA5. In many cancers, ADAR1 is overexpressed to suppress the sensing of endogenous dsRNA, thereby evading the host's immune response. By inhibiting ADAR1, this therapeutic candidate aims to induce an accumulation of unedited dsRNA, which mimics a viral infection and triggers a robust interferon-mediated innate immune response within the tumor microenvironment, potentially enhancing the efficacy of cancer immunotherapies.
